Micropropagation of Asplenium antiquum and Platycerium vassei by Spore Culture in vitro

Abstract
The present experiments were carried out to study the effects of prothallus grinding on the prothallus regeneration and sporeling development, and the effects of inorganic concentration of media on the development of prothallus and sporophytes, when culturing spores of Asplenium antiquum and Playcerium vassei on the artificial media. The effects of kinetin and auxins on the multiple shoot formation from shoot tips of young sporeling were also studied.
In Platycerium vassei, the unisexual rate of prothallus was above 70 percent, and the bisexual rate was below 30 percent. Sporophyte by fertilization of antheridia and archegonia produced from prothallus was below one percent.
Grinding by blender for 10 seconds promoted prothallus regeneration and sporeling formation, compared to using intact prothallus.
On the half strength of Knudson media, fresh weight was increased, and root development was favorable. On the media consisted of 3 parts of hyponex and 2 tryptone, fresh weight was increased, and growth of sporeling were promoted in both species. Sucrose concentration had not influenced sporeling development and growth.
Multiple shoots were obtained by culturing shoot tips of young sporeling derived from prothallus on the media supplemented with 5.0§·/§¤ kinetin. The effects of auxin on shoot tip culture were varied with kinds and concentrations, but high number of multiple shoots were obtained with 10.0§·/§¤ IBA in Asplenium antiquum and 5.0§·/§¤ IAA in Platycerium vassei.
